摘要:目的:探讨根尖牙乳头干细胞外泌体(SCAP-Exo)对牙髓干细胞(DPSCs)生物学行为的调控,重点探讨其对细胞增殖及成牙本质/成骨双向分化能力的影响。方法:基于改良酶消化法(I型胶原酶/Dispase酶消化体系)分离培养根尖牙乳头干细胞(SCAP)和DPSCs,超速离心法提取SCAP-Exo,并通过透射电镜及WB验证其特性。采用CCK-8法、茜素红S染色及RT-PCR检测SCAP-Exo对DPSCs增殖、矿化能力及成牙本质分化相关基因(DSPP)的影响。结果:SCAP-Exo呈典型双层膜囊泡结构,表达外泌体标志蛋白Alix和CD9;SCAP-Exo(50/100 μg/mL)对DPSCs增殖无显著影响(P>0.05),但显著提升矿化结节形成量(P<0.0001)并上调DSPP基因表达(P<0.0001),而对成骨相关基因OCN、Runx2无显著调控作用。结论:SCAP-Exo可通过其携带的根尖牙乳头组织的特异性生物学信号促进DPSCs成牙本质向分化,为SCAP-Exo作为一种生物制剂应用于再生性牙髓治疗提供依据。
