Abstract: Flavonoids in medicinal plants are important active substances in natural secondary metabolism, with multiple pharmacological effects such as anti-inflammatory, antioxidant, and immune regulation. They have broad application prospects in the fields of pharmaceutical preparations, functional foods, and daily chemical products. Traditional flavonoid extraction often uses single factor experiments to optimize the process, which makes it difficult to characterize the coupling interaction between extraction parameters and is prone to local optima. Moreover, the influence of process conditions on flavonoid biological activity is often overlooked. This article introduces response surface methodology to optimize the extraction process of flavonoids from medicinal plants. A second-order nonlinear regression model is constructed with ethanol concentration, solid-liquid ratio, ultrasound time, and extraction temperature as independent variables, and flavonoid yield as the response value to explore the synergistic effect of multiple factors; Simultaneously establish an in vitro inflammation model to evaluate the anti-inflammatory activity of flavonoid extracts obtained through optimized processes. The results indicate that the response surface methodology can accurately solve the global optimal extraction parameters, and the obtained flavonoid extract has a significant inhibitory effect on inflammatory factors. The research can provide theoretical reference and technological support for the efficient green extraction of flavonoids from medicinal plants and the development of natural anti-inflammatory activities.
